For many breast cancer survivors, their greatest fear comes down to one word: relapse.

Modern treatments for HER2-positive breast cancer have become remarkably effective at bringing the disease under control. But even when scans are clear, microscopic cancer cells can remain behind, creating the possibility that the disease will eventually return. HDT Bio is developing a new approach aimed at this critical period: an immune-maintenance therapy designed for the years after successful treatment.

HDT-501 is an off-the-shelf cancer vaccine designed to train the immune system’s T cells to recognize HER2-positive cancer cells and potentially maintain pressure on any cancer that remains. Unlike chemotherapy or today’s HER2-targeted drugs, the goal is to create lasting immune memory, giving the body its own surveillance system to help keep the disease under control.

Recent Phase 3 success from Merck and Moderna provides important validation that RNA-based cancer vaccines can reduce recurrence in cancer patients. HDT is taking that concept in a different direction, using its self-amplifying RNA technology and LION delivery system to create a standardized vaccine designed for repeat dosing over an extended period. HDT’s platform has already been used in more than 6,000 people, and the first HDT-501 cancer patient is targeted to be dosed in Q4 2026, with initial human immune-response data expected in Q2 2027.
